Evaluation of the biological response of Lasiodiplodia chemically modified by carboxymethylation on an animal model of hyperglycemia

The aim of this study was to test the hypoglycemic effect of b-(1→6)-D-glucan chemically modified by carboxymethylation in a group of diabetic animals. Thirty male Wistar rats were used. Diabetes was induced by administering Aloxan Monohydrate at a concentration of 125 mg/kg animal weight. Subchronically, the groups of animals received 5 and 15 mg/kg of b-(1→6)-D-glucan produced by the fungus Lasiodiplodia theobromae MMPI in submerged culture, chemically modified. The duration of the respective treatment was 28 days. Biochemical analyses were performed to quantify the levels of cholesterol, triglycerides, AST, AST, urea, creatinine and glucose, in addition to histological evaluation of the kidneys, pancreas and liver of the animals. The results obtained demonstrated that LC at a concentration of 5 mg/kg of animal weight promoted, at the end of the treatment, a reduction in fasting capillary glucose levels in rats induced to diabetes by ALX; the OGTT did not show any change in behavior in the LC5 and LC15 treatment groups when compared to the CS and MET groups; LC at a concentration of 15 mg/kg of animal weight promoted a 28% reduction in weight loss in diabetic animals when compared to diabetic saline animals; the chemical modification by carboxymethylation of lasiodiplodan caused superficial and deep damage to the liver tissue in animals in the treatment group with the lowest concentration; the serum creatinine levels of diabetic animals treated with LC were shown to be high when compared to the healthy group and normal when compared to the MET group.
